MacConkey Broth

MacConkey Broth is a selective and differential culture medium used primarily for detecting coliform bacteria in samples such as food, water, and clinical specimens. The addition of ox bile gives the medium its selective properties by inhibiting the growth of most Gram-positive bacteria. Lactose serves as the primary carbon source, and when fermented by coliform bacteria, it produces acid, causing the neutral red pH indicator to change from red to yellow.
Enriched with peptone, the medium provides the necessary nitrogenous compounds for bacterial growth. Although MacConkey Broth is a liquid medium, it is not suitable for isolating pure cultures. It is, however, highly effective for quantifying coliform populations, especially when used in conjunction with the membrane filtration method. A confirmatory test for coliform presence often involves the observation of gas production in addition to the color change.
Storage:
Store the container at 15-30°C. Prepared medium should be stored at 2-8°C.
