Yeast Extract Peptone Dextrose (YPD) Broth, also known as YEPD Broth, is a nutrient-rich, non-selective medium widely utilized for the cultivation and propagation of yeasts, particularly Saccharomyces cerevisiae. Its balanced composition supports robust yeast growth, making it an essential tool in molecular biology, genetics, and industrial microbiology applications.
 
Composition (per liter):
Peptone: 20 g
Yeast Extract: 10 g
Dextrose (Glucose): 20 g
Final pH: 6.5 ± 0.2 at 25°C

Preparation Instructions:
Suspend 50 g of YPD Broth powder in 1 liter of distilled water.
Heat if necessary to dissolve the medium completely.
Sterilize by autoclaving at 121°C for 15 minutes. 
 
Storage and Stability:
Store the dehydrated medium in a cool, dry place at 15–30°C. Once prepared, the medium should be stored at 2–8°C and used within a specified period to ensure optimal performance.
