A semi-selective medium used for the detection of Clavibacter michiganensis subsp. michiganensis on tomato seeds. This medium supports the growth of the target bacterium while inhibiting other microorganisms, making it ideal for accurate detection and isolation.

Ingredients Grams/Litre Yeast extract 2.00 Casein hydrolysate 4.00 Glucose, anhydrous 10.00 Ammonium chloride 0.15 Boric acid 1.00 Magnesium sulphate, anhydrous 1.00 Tris HCl 1.20 Agar 18.00 Final pH (at 25°C ): 7.4

Dissolve 37.3 grams of the ingredients in distilled water and adjust the volume to 1000 ml. Set the pH to 7.4.

Autoclave the solution at 121°C and 15 psi for 15 minutes.

Prepare sterile antibiotic solutions and add the following amounts per liter of medium:

28 mg nalidixic acid

100 mg cycloheximide

10 mg polymyxin B sulfate

Allow the solutions to cool down to approximately 45°C - 50°C, then add the antibiotics.

Mix gently to avoid air bubbles, and pour the medium into plates (15-20 ml per 9.0 cm plate).
