LB Broth (Lennox) – Optimized Bacterial Growth Medium
Introduced by Lennox in 1955, LB Broth (Lennox) is designed for growing and maintaining E. coli strains used in molecular microbiology. This nutritionally rich, dehydrated powder enhances the growth of recombinant E. coli strains, typically derived from K12, which can’t synthesize vitamin B and fail on nutrient-poor media. With half the sodium chloride of LB Broth (Miller)—5.0 g/L—it lets researchers fine-tune salt levels for specific strains.
How It Works
Tryptone (10.0 g/L): Delivers nitrogen, amino acids, and carbon for bacterial growth.
Yeast Extract (5.0 g/L): Supplies vitamins, trace elements, and amino acids to boost growth and plasmid yield.
Sodium Chloride (5.0 g/L): Provides electrolytes and balances osmotic pressure.
pH: 7.0 ± 0.2 at 25°C (adjusted to meet standards).
Preparation
Suspend 20.0 g in 1 L of demineralized water; mix thoroughly.
Autoclave at 121°C for 15 minutes.
Dispense into sterile containers.
