LB Agar, Miller is a nutritionally rich medium formulated for the cultivation and maintenance of Escherichia coli strains, particularly those utilized in molecular biology applications. This medium supports robust bacterial growth, making it ideal for plasmid DNA preparation, recombinant protein expression, and other genetic studies.
Composition (per liter)
Tryptone: 10 g
Yeast Extract: 5 g
Sodium Chloride (NaCl): 10 g
Agar: 15 g
The elevated NaCl concentration in the Miller formulation (10 g/L) ensures optimal osmotic conditions for E. coli strains, especially those derived from K-12 lineages.

Preparation Instructions
Dissolve 37 g of LB Agar, Miller powder in 1 liter of purified water.
Adjust the pH to 7.0 ± 0.2 as needed.
Sterilize by autoclaving at 121°C for 15 minutes.
Cool to approximately 50°C before pouring into petri dishes.
