How It Works
Agarose is a polysaccharide composed of β-D-galactopyranosyl units linked via (1,3) and (1,4) bonds to 3,6-anhydro-α-D-galactopyranosyl units. When heated with a buffer, it forms a double-helical structure that cools into a porous gel matrix. This matrix traps water and solutes, allowing DNA/RNA fragments to migrate under an electric field based on size. Adjust the agarose concentration to control pore size:
Higher Concentrations (e.g., 2%): Smaller pores for resolving smaller fragments.
Lower Concentrations (e.g., 0.8%): Larger pores for bigger fragments.
Instructions for Use
- Mix agarose powder with an appropriate buffer (e.g., TAE or TBE).
- Heat until fully dissolved, then pour into a casting tray.
